Suppressor-tRNA is an engineered tRNA molecule, of which the anticodon sequence is complementary to one of the termination codons along with other nucleotide changes. When encountering a Premature Termination Codon (PTC), the translation machinery can incorporate the aminoacylated suppressor-tRNA to add the charged amino acid to the growing peptide chain, relieving the termination effect of the PTC and producing the full-length proteins. Effect of an optimized Suppressor-tRNA on native termination codons expect to be minimal due to presence of multiple redundant stopping mechanisms at the end of normal protein translation.
